Based on the CMS paper sample on page 84 of the text, I would say that a few of the differences between Chicago Manual Style and MLA format are the use of a title page, the use of marginal annotations, and the format of the citations. While MLA format is typically used for people writing in the humanities fields, Chicago Manual Style is mainly written in the history field. In this sample, a title page is used.
